One Step Multiplex PCR for Identifications at Subspecies Level of <i>Fusobacterium nucleatum</i> and <i>Fusobacterium necrophorum</i>

Purpose: Fusobacterium nucleatum is an opportunistic pathogen involved in periodontal diseases, extraoral infections, and colorectal cancer. necrophorum causes a variety of necrotic infections. F. are classified into five two subspecies, respectively. Conventional identification methods were technically hard to distinguish each subspecies species accurately. The purpose the present study was design primers identify medically important at level, using one-step multiplex PCR. Methods: Polymerase chain reaction (PCR) designed based on partial sequences 16S ribosomal RNA (16S rRNA) gene, polymerase B (rpoB) DNA gyrase subunit (gyrB) necrophorum. Results: These able did not display cross-reactivity with representative other than Conclusion: Our developed PCR method accurate, specific, cost-effective, time-saving, worked without requiring extraction.
